"Frankie", who annoys her constantly and goes to serve in the military before applying to work in the FBI. Rizzoli has to be a mediator in the relationship of her parents, especially with Frank domineering Angela incessantly and Rizzoli having to stand up for her when crosses lines. When Frank cheats on Angela with a younger woman, Rizzoli is furious to the point of forcing the woman on her knees and nearly arresting her in front of Frank. Angela understands this meaning she doesn't deserve to be suffering those dynamics, so she leaves Frank and grows close to Detective Vince Korsak, a policeman from Brookline PD who later retires and divorces from his wife. Even after Frank guilt trips Angela into his orbit temporarily, Angela still solidifies her courage to leave him permanently, and Korsak becomes the man she turns to for love, support, and dignity.

Rizzoli struggles with being a woman on the squad, due to rampant sexism throughout the police department and constant insults and disparagement about Rizzoli, despite her persistence to the point her skills as a cop leave her missing tact she needs to stay professional even more than the men on the force. Her career is nearly destroyed when she shoots and kills a suspect in a chase, but she's ultimately cleared, despite the guilt haunting. But the worst of it is when she fights serial killer Warren Hoyt, a.k.a. "The Surgeon". In efforts to protect the women Hoyt butchers, he's briefly subdued by him, being stuck with a metal tray and pinned to the floor with scalpels through her hands. The primary target of hoyt's rage, Dr. Catherine Cordell, shoots and wounds Hoyt to save both women, and Rizzoli is lauded as a hero. When Hoyt escapes to work with an accomplice, John Stark, they kidnap Rizzoli eventually, only for her to shoot Stark dead and fire at Hoyt, paralyzing him. It takes all her willpower to not kill Hoyt in a rage to prevent from being her own translation of a monster.

Rizzoli begins to work with Dr. Maura Isles during the final manhunt for Hoyt, and when they eventually form a friendship, Isles becomes both Rizzoli's confidant and voice of reason for her extremes and doubts. Rizzoli had to return this to Isles when she finds out she's the daughter of a career serial killer, Amalthea Lank, Rizzoli constantly telling Isles to not engage Lank so as to not fall under the killer's influence to the point Isles loses herself. Isles doesn't always follow Rizzoli's instructions, but she draws lines to goad Lank enough to reveal what she is to Isles and prove they're different. Lank later dies from pancreatic cancer.

Rizzoli works with Agent Gabriel Dean as well, starting at the beginning of Hoyt's last manhunt. In spite of their combative relationship in the beginning, as they're of different ranks and statuses of law enforcement, they fall in love. When they sleep together, Rizzoli becomes pregnant with their daughter Regina, who she gives birth to after narrowly surviving a hostage situation and standoff Dean does his best to protect her from dying in. Beforehand, the two of them marry, and Dean id fully devoted as a husband and father, as well as loves her family as his own in all their tumult. He joins Rizzoli in supporting Angela and offering her their place to stay in when she sorts out how to leave the marriage she hates.

Rizzoli constantly puts her life on the line during her time on the job, which includes dangers such as gunfire, explosions, and even violently conditioned dogs. She always comes out alive, and she takes a very forward, protective, compassionate position to guard survivors of the killers she hunts with her life. Her most trusted colleague on he force is Detective Barry Frost, to whom she mutually has a dedicated partnership on the job. They tease each other, but Rizzoli is also his closest ally when his wife Alice divorces him by the time she grows into her career as a lawyer.
